The tyres in the 450 are tubeless, hence removal of the tyre for plugging small punctures is not required. The vehicle can simply be put on a paddock stand and the puncture repaired.
In case the tyre/wheel has to be removed, from the rear, the drive belt will have to be retensioned to the correct spec. We’d recommend getting this done through Ather’s service team.
